Trying to get my regal inspected. Figure the fastest thing I can do is swap on the passenger side door I have in my shed.

Which is the easiest way, unbolting the hinges from the body, or from the door?

I have some straps and an engine hoist to help me with the lifting, just wondering what will make it easier to realign.

I unbolt it from the door and then use a jack with a 2 x 4 wrapped in a towel to lift it and to adjust it, the towel keeps it from sliding and getting scratched.
Easy one man job IMO.

The engine hoist is far more stable. I’d have the hinges on the door then bolt to the car. I like to remove the striker first, line the door to the car then fine tune with the striker.

Different ways to skin that cat. Best to have a helper, though. You'll figure out how you want to do it best when you get in there. We all have done that at some point.
