A couple points to consider:
-It IS a long holiday weekend. Interrupted mail service. Seller may be out of town.
-I don't believe you can file for lost title unless the car was previously registered in YOUR name. Every state is a bit different though.
I think it might be premature to freak out. Save all your correspondence records and give it a couple more days. I wouldn't expect any help from the police since the car isn't technically stolen. It's a civil matter. Did you photocopy the title before you sent it back to the seller?
